Can a meme shape a genetic disposition?

In fact this is exactly what Nihilistic memes attempt to do.

Whereas memes emerge from genes, are extensions of genetic relationships, inherited from past, Nihilism imposes a way of thinking that results in genetic isolation - Christianity, Islam, Orthodox Jews etc.

It begins with a set of rules, ideals, and then attempts to convert them into a genetic type.
This is the inverse process of how memes actually emerge: genes in geographical isolation, producing rules, ideals, (culture) that further isolates them from world, as distinct.
Nihilistic meme begins as ideal and then imposes a harsh rule to produce cultural isolation, distinguishing itself. The ideals are contrary to anything experienced, or perceived as outside the mind. The type of man it produces, as a representation of its ideal, is usually a sickly, insane man.
A man incapable to survive outside human environments - an urban man, requiring sheltering to continue believing what he does, and distinguishing himself as such, only in relation to other humans.
In nature such a man would either perish, if he remained true to his own principles, or he would be forced to lie, contradicting his beliefs with his actions.

An example of a living meme which is the opposite of a nihilistic meme would be?

Hellenism, as the height of the pagan meme in the west.
Hinduism in the east. .
Any of the Indo-European traditions called pagan because they were practiced in rural environments when urbanization began to enforce nihilism as a method of coexisting in ms all areas.

Don't know much about their traditions, but from what I know  would say the Native Americans before the Christianized Europeans discovered them.
Although, I don't think we can call it 'living'.

The fact that most of them turned to victimhood and thrive upon it, even today, likely shows they already were infected far before any contact with the western variant.

There's a reason why you dont see "Pagan" or "Hindu" advocacy groups in the modern west.
